Efficacy for Weight Loss

Semaglutide: In the STEP 1 trial (68 weeks, 2.4 mg dose), participants lost ~15% body weight vs. 2.4% on placebo. In the 2024 SURMOUNT-5 head-to-head trial (72 weeks), it achieved 13.7% loss in adults with obesity (no diabetes).

Tirzepatide: Outperforms semaglutide in direct comparisons. SURMOUNT-1 (72 weeks, 15 mg) showed ~22.5% loss. In SURMOUNT-5, 20.2% loss (47% greater relative reduction than semaglutide), with 81% achieving ≥15% loss vs. 58% for semaglutide.

Retatrutide: Phase 2 data (48 weeks) indicate 24% average loss at higher doses (8-12 mg), surpassing both in separate studies. A 2025 systematic review ranked it highest (22% loss vs. 18% for tirzepatide, 14% for semaglutide). Ongoing phase 3 trials (e.g., TRIUMPH-1) focus on obesity without diabetes.

If retatrutide delivers 24% average loss safely, that’s insane but i wonder how tolerable it’ll be compared to sema or tirze  nausea’s already rough for some people?
